    An ELISA was performed on h Hib PS oligomers coupled to human serum albumin using the human version of this antibody (Reason, Wagner and Lucas, 1997; pmid:8975921). The human version of this antibody was used for an ELISA on Hib PS-poly-L-lysine. Furthermore, an infant rat assay was used to evaluate in vivo protection of this antibody, Five- to 7-day-old rats received a subcutaneous injection of either purified IgGl or IgG2 anti-Hib PS antibodies, purified MAb CA4, or, as a negative control, a human IgG2 myeloma protein. Eighteen to 24 h later, blood was taken and assayed quantitatively on chocolate agar for the presence of Hib organisms. Additionally, this antibody was evaluated by an ELISA for rheumatoid factor activity and for binding to histones, chromatin, and denatured DNA (Lucas, Larrick and Reason, 1994; pmid:8063404).

    This antibody was raised by immunizing a 43-year-old white male using a subcutaneous injection of 40 mg of Hib PS vaccine. Five days later, mononuclear cells (MNC) were isolated from 60 ml of peripheral blood with Lymphoprep. Two cell populations were used for RNA isolation: unfractionated MNC and a HibId-1-enriched population, isolated with magnetic beads.
