This antibody immunoprecipitated a 32 kD polypeptide from lysates of the ovarian tumor cell line SKOV3. Cell surface immunofluorescence determinations using the fluorescence activated cell sorter revealed that this antibody stain ovarian or cervix carcinoma tumor cell lines (PMID: 7756578). Immunoperoxidase staining with this antibody of nonpermeabilized cryostat sections of freshly obtained or cryopreserved ovarian carcinoma specimens and human ovarian tumor xenografts demonstrated strong reactivity of these specimens. This antibody mediated complement-dependent cytotoxicity and identified a 32 Kd band in western blotting and immunoprecipitation experiments conducted on surface labelled SKOV3 cells. The association constant for the IgM version of the antibody was determined at 2.3 x 10^(10) M^(-1) (PMID: 2032733). The biodistribution of 111In-labeled IgM conjugate of this antibody was studied after i.v. or i.p. administration in nude mice bearing an s.c. xenograft or peritoneal tumor lumps of a human colorectal carcinoma (SW620). The i.p. administration of intact, specific radiolabeled IgM provides prolonged retention of radioactivity in tumor, low normal tissue uptake, a long peritoneal residence time, and very limited spillover of IgM into the circulation (PMID:7493338).
