    The specificity of this antibody for murine CD226 has been confirmed in flow cytometric analysis using 293 cells transfected with CD226 cDNA and untransfected 293 cells (Dardalhon et al, 2005). This antibody has been used in flow cytometry to identify the surface expression pattern of CD226, which is predominantly confined to T cells (Dardalhon et al, 2005; Yasuma et al, 2016), and to detect NK cell CD226 expression and apoptosis (Huang et al, 2011). Immunisation of mice with this antibody inhibits antigen-specific T cell expansion (Dardalhon et al, 2005). Additionally, treatment of experimental autoimmune encephalomyelitis (EAE) mice with this antibody delays the onset and reduces the severity of Th1 cell-mediated autoimmune disease (Dardalhon et al, 2005).

Figure 1. Western blot analysis of CD226 using anti-CD226 antibody (A01094-2). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30ug of sample under reducing conditions. Lane 1: rat spleen tissue lysates, Lane 2: mouse spleen tissue lysates, Lane 3: mouse thymus tissue lysates, Lane 4: mouse Raw264.7 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD226 antigen affinity purified polyclonal antibody (Catalog # A01094-2) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for CD226 at approximately 70KD. The expected band size for CD226 is at 70KD.
