- Antibody SpecificityThis antibody is specific for human CD73, which is a 70 kD glycophosphatidylinositol (GPI)-linked 5-nucleotidase. This antibody does not show cross reactivity with mouse CD73.
- Application Supplier NoteThe enzymatic activity of CD73 causes hydrolysis of extracellular nucleotides into membrane permeable nucleosides. This antibody promotes cell-surface clustering of CD73 followed by internalization, thus inhibiting the ability of circulating tumor cells to extravasate and colonize, leading to inhibition of metastasis. This antibody inhibited development of metastasis in a spontaneous animal model of human metastatic breast cancer. Flow cytometry analysis showed that anti-CD73 AD2 mAb (AD2 mAb) recognized CD73 on the human breast cancer cell lines LM3 and MDA-MB-231, but failed to detect the murine CD73 variant expressed on isolated mouse mononuclear hematopoietic cells(PMID: 24043904). This antibody was also used to study the expression of CD73 in normal mammary gland and breast carcinoma cells using immunohistochemistry (PMID: 1989648). mAb AD2 was also used in the immunofluorescence staining for detection of human CD73 in the thymus, peripheral blood, spleen, and lymph node in transgenic mice (PMID: 9045870). Bispecific antibody generated using 1E9 and mAb AD2 is reported to inhibit the enzymatic activity of CD73 as well as promotes internalization of CD73.
