- Application Supplier NoteThe antibody reacted with conformational epitopes expressed by partially dissociated oligomers, but not with monomer as elucidated by Western blot analysis. Ascites fluid containing the antibody showed the inhibitory activity against P. gingivalis to saliva-coated hydroxyapatite beads, an in vitro model for the pellicle-coated tooth surface. For example, 400-fold diluted antibody inhibited the binding of the bacteria onto bead surfaces of 29.5% (Koh et al., 2009; PMID: 20157608). The specificity of this antibody (IgG2b) was confirmed by ELISA analysis. The original antibody detected FimA by western blot. The antibody significantly inhibited the binding of P. gingivalis to the surface of saliva-coated hydroxyapatite (SHA) beads by 49.79 +/- 5.0%, when 50 ng/mL antibody was used (P < 0.01). The antibody could be used as a basis for passive immunization against P. gingivalis-mediated periodontitis (Koh et al., 2011; PMID: 21223367).
