- Application Supplier NoteGelsolin is a multifunctional actin-binding protein that controls cellular actin organization and migration. The original VHH version of this antibody binds gelsolin with a Kd of ~5 nM irrespective of calcium and completely blocks its interaction with G-actin. The specificity of the antibody towards gelsolin was confirmed with ELISA. GsnVHH 13 expressed as intrabodies immunoprecipitate endogenous gelsolin. His-tagged GsnVHH 13 can be used to identify gelsolin in a western blot assay. This antibody can trace gelsolin in membrane ruffles of EGF-stimulated MCF-7 cells and delay cellmigration without affecting F-actin severing/capping oractin nucleation activities by gelsolin as demonstrated by immunofluorescense staining. It was also reported that free gelsolin is targeted to the nucleus by nuclear localizationsequence (NLS)-GsnVHH 11 in HeLa cells (PMID:20140750). Hereditary gelsolin amyloidosis is an autosomal dominantly inherited amyloid disorder caused by a point mutation in the GSN gene resulting in disturbed calcium binding and improper folding of the second gelsolin domain (G2) and rendering the mutant plasma gelsolin susceptible to a proteolytic cascade. This antibody was also used as a molecular chaperone to protect mutant plasma gelsolin from furin proteolysis in the trans-Golgi network (PMID: 25601851). This antibody was also used in the study of structure, dynamics and proteotoxicity of the Finnish-type amyloidogenic gelsolin variant (PMID: 30625383).
