- Antibody SpecificityThe antibody is specific for JEV. It binds a quaternary epitope that span across three adjacent envelope proteins. The antibody does not cross react with E protein from dengue (DENV1 and DENV2), Zika (ZIKV) and yellow fever (YFV) viruses.
- Application Supplier NoteThe specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody was used in indirect immunofluoresenceThe antibody could neutralize the virus in mice (Zhang et al., 1989; PMID: 2557383). Surface plasmon resonance experiments showed that the antibody binds to the JEV rE with a high affinity of 2.9nM. The antibody could block attachment of the virus to its receptor and also prevent fusion of the virus. Cell-based neutralizing assays (in Vero cells) revealed that the antibody has strong neutralizing activities: a 50% neutralizing concentration value of 1.2 ng ml-1 against JEV (P3 strain). The antibody protected 100% of mice from lethal infection of JEV and led to complete clearance of JEV from the brains of mice, indicating its potential as a therapeutic agent. The structure of the Fab fragment of the antibody was determined by X-ray and of the Fab in complex with JEV by Cryo-EM (Qiu et al., 2018; PMID: 29379207).
