- Application Supplier NoteWhile screening for antibodies against E1 Glycoprotein, an ELISA was performed on peptide ITGHRMAWDMMMNWS using the human version of this antibody. Further the ability of this antibody to neutralize retrovirus pseudo particles (HCVpp) of H77, BE-11, ED43, SA13, HK, JFH1 and J6CF strains bearing HCV envelope glycoproteins was assessed. To do this the pseudo particles were incubated with the human version of this antibody and then added to huh-7 cells. These huh-7 cells were then analyzed using flow cytometry. This antibody could successfully neutralize HCVpp bearing the envelope glycoproteins of genotypes 1a, 1b, 4a, 5a, and 6a and less strongly neutralize HCVpp bearing the envelope glycoproteins of genotype 2a. Genotype 3a was not neutralized (Meunier et al, 2008; pmid:17977972). To study the ability of peptides in inhibiting this antibody binding to E1E2, the peptides and the human version of this antibody were mixed at the specified ratio and incubated before adding to ELISA microwells coated with lectin-captured E1E2. Further X-ray crystallography was performed on the Fab fragment of this antibody bound to its antigen to elucidate the structure of the bound antibody (Kong et al, 2015; pmid:26135247). To assess the function of E glycoprotein in virus entry and assembly, immunofluorescence was performed on Huh-7 cells electroporated with wild-type or mutant RNA using the human version of this antibody (Haddad et al, 2017; pmid:28179528). To study of the C-terminal part of the hepatitis C virus E1 ectodomain a western blot was performed on lysed gug-7 cells using the human version of this antibody (Moustafa et al, 2018; pmid:30068644). To detect bound E1 glycoprotein flow cytometry was performed using the human version of this antibody on 293T/CLDN1 or Huh7.5.1 cells infected with HCV pseudo particles (Krieger et al, 2010; pmid:20069648). In research towards a vaccine against hepatitis C, an ELISA was performed on H77c E1E2 using the human version of this antibody (Wong et al, 2014; pmid:25275133).
