- Application Supplier NoteThe binding specificity of this antibody towards the E1E2 complex was evaluated using a competition ELISA using antibodies specific for AR1, AR2, or AR3. This antibody does not particularly interfere with the binding of E1E2 complex with CD81. This antibody is reported to have an exceptionally broad neutralizing activity toward diverse HCV genotypes and protects against heterologous HCV challenge in a small animal model. It was reported to neutralizing various strains of HCV belonging to genotype 1a, 1b, 2a, 3a, 4a, 5a and 6a. In an immunoprecipitation assay the human IgG1 version of this antibody was able to precipitate wild-type E1E2 complex but not the E1E2 mutant xN196/305A (PMID: 22492964). This antibody alone and in combination with epigallocatechin gallate (EGCG) was used in the in vitro (using a HCV cell culture system) and in vivo (using a humanized liver mouse model capable of supporting HCV replication) study for protection against the establishment of HCV genotype 1a infection (PMID:26389583). This antibody was also used in a flow cytometric study where a library of alanine scanning mutants comprising the entirety of E1E2 (555 residues) was created and probed for binding with the co-receptor CD81 and the antibody to E1E2 complex to evaluate the role of each residue in the glycoproteins (PMID: 29253863).
