    This antibody did not compete with IL-15 for binding to its receptor but potently interfered with the assembly of the IL-15 receptor alpha, beta, gamma complex. This antibody could bind recombinant human IL-15 in an ELISA. In vitro, this antibody effectively blocked IL-15-induced T cell proliferation and monocyte TNF-alpha release. In a human psoriasis xenograft model, antibody 146B7 reduced the severity of psoriasis, as measured by epidermal thickness, grade of parakeratosis, and numbers of inflammatory cells and cycling keratinocytes (PMID: 14617758). IL-15 is implicated in coeliac disease. A randomized, double-blind, placebo-controlled, parallel-group, phase 2a trial was done at three clinical sites in Finland to investigate the effects of AMG 714 in patients with coeliac disease who underwent gluten challenge (PMID: 31494096). The binding of this antibody to IL-15 inhibits the interaction of IL-15 with the IL-2Rbeta and common gamma chain of the IL-15 receptor complex, but not with the IL-15Ralpha chain (PMID: 36083248). Upon administration, ordesekimab binds to and neutralizes IL-15, thereby preventing IL-15-mediated pro-inflammatory signaling. By inhibiting IL-15-mediated immune responses, ordesekimab decreases natural killer (NK) cell activation and proliferation, reduces T-cell infiltration, increases T-cell apoptosis, and may prevent the growth of IL-15-driven cancer cells (National Cancer Institute, NCI Drug Dictionary).
