Lewis Y (Ley) is a blood group antigen expressed at high levels on the surface of epithelial tumors, including small cell lung cancer.

The murine 3S193 showed high specificity for Ley in ELISA tests with synthetic Ley and Ley-containing glycoproteins and glycolipids and also reacted strongly in rosetting assays and cytotoxic tests with Ley-expressing cells (Scott et al, 2000). Murine 3S193 was humanised and in antibody-dependent cell-mediated cytotoxicity assays displayed 100-fold greater activity (Kitamura et al, 1994). 3S193 has been used to screen tumour samples for Ley expression using immunohistochemistry (Krug et al, 2007). A crystal structure of the humanised antibody has been obtained to 1.9 A resolution (Ramsland et al, 2004).
