- Application Supplier NoteThis antibody binds to the extracellular domain of LMP1, which was confirmed with ELISA. The ability of the expressed HLEAFab to specifically bind to native LMP1 on the HNE2/LMP1 cell surface was further confirmed by Cell ELISA. The binding was also confirmed using flow cytometry and immunofluorescence staining. An immunoconjugate fof HLEAFab and mitomycin C (HLEAFab-MMC) inhibits proliferation and induced apoptosis in nasopharyngeal carcinoma (NPC) cell line HNE2/LMP1 in vitro and is also found to show inhibition of growth of NPC xenografts in nude mice (PMID: 22169768). HLEAFab works synergistically with mitomycin C to inhibit growth of nasopharyngeal carcinoma growth in vivo through inducing apoptosis and downregulating vascular endothelial growth factor (VEGF). Immunohistochemical staining showed that VEGF expression in NPC xenografts was significantly inhibited in the combination group compared to the Fab (PMID: 22408448). HLEAFab is recommended to be used to detect LMP1 expression in NPC tissues using immunohistochemical staining. Positive staining of LMP1 was observed in the membrane and cytoplasm of NPC cells (PMID: 23760364).
