- Antibody SpecificityThis antibody specifically binds hen egg-white lysozyme.
- Application Supplier NoteLysozyme is an enzyme that degrades polysacchardies of various types of bacterial cell walls. It is observed that lysozyme bound D1.3 still retains its enzymatic activity. This is attributed to the fact that D1.3 binds lysozyme at a site remote from the catalytic site and that there is no significant conformational change caused by the binding (PMID: 3614217). The three dimension structure of a complex between the Fab from antibody D1.3 and its antigen, HEL, has been determined at 6 and at 2.8 a resolution (PMID: 2426778, 3965976). Sixteen residues of lysozyme make contact with 17 residues of the antibody. The antigenic determinant is topographic including lysozyme residues 18, 19, 21-25, 27, 116-121, 124, and 129. All six complementarity determining regions of the antibody contact the antigen, and in addition, two residues from framework segments of V H and V L (Amit et al., 1986; PMID: 2441250). D1.3 binds native lysozyme in an ELISA (PMID: 2448386). Humanized version of this antibody is also available (PMID: 1560463, 2451287). Flow cytometry was used to find the fine difference in affinity of D1.3 (scFv) and mutant M3 clone expressed in a yeast surface display (PMID:10662486).
