    This antribody is specific for the G2 domain of nidogen-1. It binds in such a way that it is able to stop the binding of nidogen to laminin. Nidogen-1 is a sulfated glycoprotein widely distributed in basement membranes and tightly associated with laminin.

    The binding of the mouse version of this antibody to nidogen-1 was proven by ELISA. Further, high binding affinity of the mouse version of this antibody to nidogen-1 was demonstrated using surface plasmon resonance (Ries et al, 2001; pmid:11589703). While researching nidogen in healthy human cartilage and in late-stage osteoarthritis cartilage an inhibition assay was preformed using the mouse version of this antibody. In short, wells were coated with either nidogen-1 or nidogen-2 to which this antibody was added. Then, chondrocytes were added and their adhesion to the nidogen was tested. 30-40% of chondrocytes showed adhesion to nidogen-1. (Kruegel et al, 2008; pmid:18438862). Tissue sections from Female New Mexico Research Institute (NMRI) mice were stained for immunohistochemistry using this antibody as a positive control. This experiment was preformed to assess the binding between laminin and nidogen (Gersdorff et al, 2005; pmid:15824114). Tissue sections from aborted human embryos were stained with the mouse version of this antibody to be used for immunohistochemistry (Miosge et al, 2001; pmid:12005023). In research on osteoarthritis multiple experiments were preformed using the mouse version of this antibody. First, tissues from DDR-1 KO mice were stained for immunohistochemistry using the mouse version of this antibody. Second, immunoblotting was preformed on proteins extracted from DDR-1 KO mice. Third, this antibody was used to stain cell cultures taken from TMJ cartilage tissue of 9-week-old DDR-1 KO mice and their WT littermates. Finally, the cell cultures taken from TMJ cartilage tissue of 9-week-old DDR-1 KO mice and their WT littermates were used for flow cytometry and stained with this antibody (Schminke et al, 2014; pmid:23912900). In research on nidogen-2, the mouse version of this antibody was used to immunolocalize nidogen-1 in both immunofluorescence and immunohistochemistry on cells and tissues from mice with a mutated nidogen-2 gene (Schymeinsky et al, 2002; pmid:12215539).
