- Application Supplier NoteThe original antibody (mouse IgM) was generated and screened via ELISA against for gangliosides of interest. This antibody was used for tumor recognition in IHC, showing a strong positive reaction in ductal infiltrating breast carcinoma, breast cancer metastasis in lymph nodes, and breast adenosis tissue, with no cross-reactivity to other malignant or benign tumors. Additionally, it was used in in vivo experiments with Balb/c mice to stimulate immune responses, generating a significant IgG-type anti-idiotypic (Ab2) response, which increased when conjugated to KLH and administered with adjuvants. This antibody also produced an anti-anti-idiotypic (Ab1) response, demonstrating its ability to connect to both T and B cell networks and generate a specific antigen-independent immune response (EP0657471B1). This antibody was used as a positive control in an in vitro and in vivo study (Carr et al., 2002; PMID: 12573110). A chimeric(humanized; human IgG1) variant of this antibody was created (chP3), and the crystal structure of its Fab was solved (Talavera et al., 2009; PMID: 19748674).
