    The antibody is specific for PD-1. PD-1 is a type I membrane protein with 288 amino acids, and is one of the main known immune checkpoints. PD-1 is expressed in activated T lymphocytes, and it binds ligands PD-L1 and PD-L2.

    The specificity of the original format of the antibody was confirmed by ELISA analysis. The original format and the humanized version of the antibody were capable of blocking the binding of PD-1 and PD-L1. The antibody was able to activate the T cells under in vitro culture conditions and promote the production of IFN-gamma, thereby enhancing their T cell immune function. The binding affinity of the murine and humanized versions of the antibody to PD-1 were measured by surface plasmon resonance (Kd= 6.74x10 -10 M and 8.07x10 -10 M respectively). The antibody blocked the interactions between PD-1 and PD-L1. The antibody inhibited tumor growth in NCG mouse models of non-small cell lung cancer and colorectal cancer tumors (CN107043420B).
