- Antibody SpecificityThis antibody specifically binds the amino acids 318-510 in the S1 domain of the SARS-CoV Spike protein (Urbani strain).
- Application Supplier NoteThis antibody inhibits the association of SARS-CoV s protein to the angiotensin-converting enzyme 2 (ACE2) receptor by binding to specific amino acids on the S1 domain of the spike protein of the virus. The originally characterized antibody had a neutralizing titer 200TCID50 of 12.5 microg/ml (Coughlin et at.,2006). When incubated with S12-510- Fc recombinant protein it failed to block the interaction of this protein with the cellular receptor but inhibited a post-binding event and prevented viral entry. Combination of this antibody with another antibody 5D3, that binds to a different epitope resulted in increased inhibition of virus. The ability of mAb 4D4 to more efficiently inhibit viral entry when added post virus binding suggests that the S protein might undergo a conformational change following binding to ACE2 leading to unmasking of the epitope recognized by the 4D4 antibody. Simultaneously, 4D4 was also used in a standardentry inhibition assay as a positive control for determining the degree of inhibition of pseudotyped virus entry. (Coughlin et at.,2009). This antibody 4D4 showed about 50% binding to Sin845 S1 protein relative to their binding to Urbani S1 protein. GZ0402-S1 protein showed minimal of 57% binding to HmAbs 4D4 (Elshabrawy et al., 2012).
