B-Xpander™ is a potent enhanced Multimeric human CD40L cytokine, for B cell activation, B cell expansion (proliferation) and cell therapy applications. In-house production using a proprietary protocol and a characterized and certified CHO cell line allows the supply of the most active Multimeric human CD40L proteins in GMP-like format. Interest in B cells has recently increased in immunotherapies using Engineered B cells or Tumor-Infiltrating Lymphocytes (TILs). However, current methods for B cell expansion are inefficient, time consuming and can lead to insufficient B cell numbers and persistent T cell exhaustion. Our B-Xpander™ protein improves current protocols for B cell stimulation by mimicking the natural interaction between CD40L and CD40 molecules on B cells. CD40 ligand (CD40L) expressed by activated T cells engages CD40 expressed by B cells and APCs. CD40L can provide signals to B cells that induce proliferation, immunoglobulin class switching and antibody secretion. CD40L has been used recently in Immunotherapy to activate tumor-infiltrated lymphocytes (TIL). MultimericCD40L™ is a high-activity construct in which two trimeric CD40 ligands are artificially linked via the collagen domain of ACRP30. This construct very effectively simulates the natural membrane-assisted aggregation of CD40L in vivo. It provides a simple and equally potent alternative to [CD40L+enhancer] combinations. MultimericCD40L™ has been shown to be a potent tool for B cell expansion. It also has big potential as a growth factor for tumor-infiltrating lymphocytes (TILs), which have been shown to be important for T cell therapy. The CD40L (human) (multimeric) (rec.) (Animal Free) has been produced in CHO in suspension using animal-component free medium and certified CHO cells.
