Hck-IN-1, a selective Nef-dependent Hck inhibitor, blocks the kinase activity of the Nef:Hck complex in vitro with an IC50 value of 2.8 microM, whereas its activity against Hck alone is >20 microM, also blocks wild-type HIV-1 replication with an IC50 value in the 100-300 nM range.
