  • Scientific Description
    This gene encodes a lymphokine involved in cell-mediated immunity, immunoregulation, and inflammation. It plays a role in the regulation of macrophage function in host defense through the suppression of anti-inflammatory effects of glucocorticoids. This lymphokine and the JAB1 protein form a complex in the cytosol near the peripheral plasma membrane, which may indicate an additional role in integrin signaling pathways. [provided by RefSeq]

Figure 1. Western blot analysis of MIF using anti-MIF antibody (PB9274). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions. Lane 1: human Jurkat whole cell lysates, Lane 2: human K562 whole cell lysates, Lane 3: human Hela whole cell lysates, Lane 4: human HepG2 whole cell lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-MIF antigen affinity purified polyclonal antibody (Catalog # PB9274) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for MIF at approximately 12 kDa. The expected band size for MIF is at 12 kDa.

Western blot analysis of GeneTex's IgY fraction of Chicken-anti-Human MIF polyclonal antibody shows the detection of 100 μg of recombinant MIF present in a lysate. Similar detection of MIF will occur when human serum is analyzed. In lane 1 no reaction is observed in the control whereas lane 2 shows a single band at 12.3 kDa. A 4-20% gradient gel was used to separate the proteins by SDS-PAGE. The protein was transferred to nitro-cellulose using standard methods. After blocking the membrane was probed with the primary antibody for 1 h at room temperature followed by washes and reaction with a 1:5,000 dilution of IRDye800 conjugated Gt-a-Chicken Rabbit IgG [H&L] for 1 h at room temperature. LICOR's OdysseyR Infrared Imaging System was used to scan and process the image. Other detection systems will yield similar results.
