FACS analysis of cells with MAb to OPG (human) (GTX18051). HEK 293T cells (5x105) were mock transfected (thin line) or transfected with an expression plasmid enabling surface expression of the TNFR homology domain of human OPG (thick line). Cells were incubated on ice for 30 min. in 50microl FACS buffer (PBS, 5% fetal calf serum, 0.02% azide) containing 1microg/ml of MAb to OPG (human) (Bony-1) (GTX18051). After washing in FACS buffer, RPE-conjugated antibody to rat IgG was added. Cells were incubated on ice for 30 min, washed and then analyzed by flow cytometry.

- Scientific DescriptionOsteoprotegerin (OPG) is a member of the TNF receptor superfamily. This protein is an osteoblast-secreted decoy receptor that functions as a negative regulator of bone resorption. This protein specifically binds to its ligand, osteoprotegerin ligand (TNFSF11/OPGL), both of which are key extracellular regulators of osteoclast development. Studies of the mouse counterpart also suggest this protein and its ligand play a role in lymph-node organogenesis and vascular calcification. Alternatively spliced transcript variants of the gene have been reported, but their full length nature has not been determined.
