
WB analysis of NIH3T3 stimulated with 50ng/mL PDGF-BB for 10 mins (Lane 2-5) using GTX25443 PDGF Receptor alpha + beta (phospho Tyr572/Tyr574) antibody prior incubated with the non-phosphopeptide corresponding to the phosphopeptide immunogen (Lane 3), a generic phosphotyrosine containing peptide(Lane 4), or the phosphopeptide immunogen (Lane 5). The data show that only the immunogen phosphopeptide blocks the signal, demonstrating the specificity of the antibody.

- Scientific DescriptionPlatelet-derived growth factor receptor (PDGFR) is a transmembrane glycoprotein of 170-185 kDa which undergoes homo- or heterodimerization into complexes of alpha and beta subunits upon ligand binding, depending on the isoform of PDGF (PDGF-AA, -BB or -AB) that binds. The phosphorylation of tyrosine residues in the now activated receptor can control multiple signaling events such as actin reorganization, transcription, cell growth, migration and differentiation. PDGFR alpha tyrosines 572 and 574 (579 and 581 in PDGFR beta) are autophosphorylated in the activated receptor, and bind and activate Src family kinases.
