When diluting test samples, first note the dilution factor for future data analysis. For example, a human plasma sample was diluted 1:10 (50 microL plasma sample + 450 microL plasma sample diluent) and yielded a standard curve derived analyte concentration of 50 ng/mL from the ELISA assay. This concentration should then be multiplied by 10 (the dilution factor) to obtain the actual concentration value of 500 ng/mL in the undiluted plasma sample.

Plasma Sample Diluent is formulated specifically for use with plasma samples being tested in an antigen-down ELISA format. This novel mammalian-protein-buffered formulation provides an assay environment that minimizes non-specific binding while maintaining proper ionic strength and pH for efficient antibody-antigen interaction. Plasma Sample Diluent also inhibits thrombin activity (clotting) and complement activity during the plasma sample incubation period.
