    Protein. Signal peptide and extracellular domain of mouse DLL4 (aa 1-532) is fused at the C-terminus to the Fc portion of human IgG1. Source: HEK 293 cells. Endotoxin content: <0.1EU/microg purified protein (LAL test; Lonza). Liquid. 0.2microm-filtered solution in PBS. Purity: >90% (SDS-PAGE). The Notch ligand delta-like protein 4 (DLL4) is expressed highly and selectively within the arterial endothelium and has been shown to function as a ligand for Notch1 and Notch4. It is induced by VEGF as a negative feedback regulator and acts to prevent overexuberant angiogenic sprouting, pro-moting the timely formation of a well differentiated vascular network. DLL4-Notch1 signaling regulates the formation of appropriate numbers of tip cells to control vessel sprouting and branching in the mouse retina. - The Notch ligand delta-like protein 4 (DLL4) is expressed highly and selectively within the arterial endothelium and has been shown to function as a ligand for Notch1 and Notch4. Mouse NLRP1b (mouse) (full-length and UFA-CARD fragment) is detected in Raw264.7 cells using NLRP1b, mAb (2A12) (Prod. No. AG-20B-0084). Figure 1: Immunoblot analysis of beta-tubulin protein glutamylation using anti-beta-Tubulin, mAb (AXO45) (Prod. No. AG-20B-0085). Method: HEK-293T cells are grown in standard culture conditions, transfected with plasmids expressing the different glutamylases, TTLL4 and TTLL6 and are run on a 10% SDS-PAGE (specific protocol to separate alpha- and beta-tubulin (Magiera & Janke, 2013)). The proteins are transferred to a nitrocellulose membrane and detected by standard immunoblot protocol using the AXO45 (1:1000) in PBS containing 0.1% Tween-20 for washing steps and 2.5% fat free milk for antibody incubation. In all the cell extracts, AXO45 specifically detects beta-tubulin, with no cross-reactivity to alpha-tubulin. Glutamylation added by either TTLL4 or TTLL6 appears to not affect this detection.
