WB (peptide competition) analysis of human recombinant Tau stimulated with GSK-3 beta (1 microg per microg Tau) for 45 minutes using GTX24862 Tau (phospho Ser422) antibody prior incubated with the non-phosphopeptide corresponding to the non-phosphopeptide immunogen (Lane 2), a generic phosphoserine-containing peptide (Lane 3), or the phosphopeptide immunogen (Lane 4) control. The data show that only the immunogen phosphopeptide blocks the signal, demonstrating the specificity of the antibody. The membrane treated with phosphatase (Lane 5) eliminates the signal further verifying that the antibody is phospho-specific.

    This gene encodes the microtubule-associated protein tau (MAPT) whose transcript undergoes complex, regulated alternative splicing, giving rise to several mRNA species. MAPT transcripts are differentially expressed in the nervous system, depending on stage of neuronal maturation and neuron type. MAPT gene mutations have been associated with several neurodegenerative disorders such as Alzheimers disease, Picks disease, frontotemporal dementia, cortico-basal degeneration and progressive supranuclear palsy.
