- Scientific DescriptionAnti-4R-Tau pAb - prepared from rabbits immunized with synthetic N279D 4R-tau peptide [VQIIDKKLDLSNVQSKC] - recognizes 4R tau isoforms, regardless of deamidation as occurs extensively in AD brains. Validated for western blot and IHC(p), this isoform-specific tau antibody is useful for immunohistochemical and biochemical studies of tau species in diverse neurodegenerative diseases. Tau is a microtubule-associated protein that assembles into insoluble, filamentous, and hyperphosphorylated inclusions in many neurodegenerative diseases. The adult human brain expresses 6 distinct splice forms divided into two groups depending on the inclusion of exon 10 to generate either 3-repeat (3R) or 4-repeat (4R) isoforms. Distinct tau isoforms accumulate in diseased brains: all 6 isoforms in AD, 3R tau in Picks disease, and 4R tau in PSP and CBD. Therefore, isoform-specific tau antibodies are useful tools for immunohistochemical and biochemical studies of tau species in diseased brains. Tau deposited in AD is extensively deamidated at N279. This anti-4R tau pAb specifically recognizes 4R tau isoforms regardless of deamidation and strongly stained tau in AD brain.
