    The binding of this antibody to CD112 or Nectin-2 expressed on the surface of CHO cells was determined using whole cell ELISA. The cross reactivity of this antibody for cynomolgus monkey CD112 expressed on CHO-K1 cells was checked using flow cytometry. This antibody shows good ADCC activity against OV-90, MDA-MB-231, NCI-H1299, Du145 and U937 cell lines. The in-vivo anti-tumor activity of this antibody was tested in nude mice models transplanted with OV-90 human ovarian cancer cell line. This antibody showed T/C (Treatment/Control) value of 34%. The in-vivo activity was also checked in SCID mouse models for treating lung metastasis with MDA-MB-231 human breast cancer cell line (US20100008928).

Figure 1. Western blot analysis of Nectin 2/NECTIN2 using anti-Nectin 2/NECTIN2 antibody (A08081-2). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human K562 whole cell lysates, Lane 2: human HEK293 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Nectin 2/NECTIN2 antigen affinity purified polyclonal antibody (Catalog # A08081-2) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Nectin 2/NECTIN2 at approximately 70-80KD. The expected band size for Nectin 2/NECTIN2 is at 58KD.
