The binding specificity of this antibody has been confirmed both in ELISA analysis using recombinant murine CD155 (mCD155), and in flow cytometric analysis using HEK293 cells transiently expressing mCD155 (Maier et al, 2007). This antibody has been used in flow cytometry to assess mCD155 expression on lymphocytes and dendritic cells (Maier et al, 2007; Seth et al, 2011). This antibody blocks the interaction of CD155 with its ligands CD96 and CD226 (Qiu et al, 2010). When administered to WT mice, antibody-induced disruption of the CD155-CD226 interaction has been shown to cause CD226 upregulation on CD4+ T cells and CD8+ T cells (Seth et al, 2011). Administration of this antibody has also been shown to stimulate the loss of mature, memory-like CD8+ T cells from the thymus (Seth et al, 2011; Georgiev et al, 2016).

Figure 1. Western blot analysis of Poliovirus Receptor/PVR using anti-Poliovirus Receptor/PVR antibody (A00664-3). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30ug of sample under reducing conditions. Lane 1: rat C6 whole cell lysates, Lane 2: rat PC-12 whole cell lysates, Lane 3: mouse NIH/3T3 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Poliovirus Receptor/PVR antigen affinity purified polyclonal antibody (Catalog # A00664-3) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Poliovirus Receptor/PVR at approximately 60-80KD. The expected band size for Poliovirus Receptor/PVR is at 60-80KD.
