    Human CD155 (Polio Virus Receptor, PVR, Necl-5) is a 70kd type I Ig superfamily molecule. It is involved in formation of intracellular junctions between epithelial cells. Its ligands include CD226 (DNAM-1), and CD96 (TACTILE). CD155 expression by tumor has been shown to be upregulated by nitric oxide. A soluble version of CD155 has been shown to exist. High CD155 expression has recently been exploited to use engineered poliovirus to treat glioblastoma. - Monoclonal Antibody. Binds to human cell surface CD155 on U-937 cells, and binds to recombinant human CD155 in EIA. Applications: ELISA, FACS. Isotype: Mouse IgG1kappa. Clone: ANC2B2. Figure 1. Western blot analysis of Poliovirus Receptor/PVR using anti-Poliovirus Receptor/PVR antibody (A00664-2). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30ug of sample under reducing conditions. Lane 1: human Hela whole cell lysates, Lane 2: human A549 whole cell lysates, Lane 3: human HT1080 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Poliovirus Receptor/PVR antigen affinity purified polyclonal antibody (Catalog # A00664-2) at 0.25 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for Poliovirus Receptor/PVR at approximately 70-80KD. The expected band size for Poliovirus Receptor/PVR is at 70-80KD.
