The specificity of this antibody for alpha5 integrin has been confirmed by ELISA using immobilised alpha5beta1 integrin (Clark et al, 2005). This antibody is conformation-dependent, and is able to convert alpha5beta1 integrin into a ligand-competent form and promote fibronectin binding. In a K562-cell adhesion assay, this antibody has been shown to stimulate alpha5beta1 integrin to bind fibronectin, and, by extension, promote cell adhesion (Clark et al, 2005). The ability of this antibody to stimulate alpha5beta1 integrin function has been confirmed in solid-phase ligand-binding assays (Clark et al, 2005). This antibody has been used to specifically stain integrin localised to fibrillar adhesions in indirect immunofluorescence assays (Clark et al, 2005). It has also been used as an activation reporter to allow the early endocytic steps of active alpha5beta1 integrin in endocytic cells to be assessed in fluorescent confocal analysis and immunofluorescence analysis (Valdembri et al, 2009).
