    HMD4-2 recognizes murine Dll4, one of the five major ligands of the Notch signalling pathway. DLL4 signalling has been shown to play a role in the angiogenesis of clear-cell renal tumours, and pancreatic, bladder and colonic cancer.

    This antibody can be used to investigate the role of the signalling pathways related to the Notch receptors and its ligands. The antibody blocks Notch1-Fc and Notch4-Fc binding to mouse Dll4-expressing cells in a dose-dependent manner and does not bind to cells expressing other mouse Notch ligands, such as Dll1, Jagged1, or Jagged2 (Sekine et al., 2009; 19181931). HMD4-2 was demonstrated as an effective infibitor able to neutralize the Dll4-dependent Notch signalling (Sekine et al., 2009; 19181931). It was used to block Dll4-mediated Notch signaling in LDL-receptor-deficient (Ldlr-/-) mice fed a high-fat, high-cholesterol diet, a model that produces atherosclerosis and metabolic disturbances resembling those seen in the cardiometabolic syndrome in order to test the hypothesis in vivo that Dll4 contributes to the pathogenesis of the cardiometabolic syndrome (Fukuda et al., 2012; pmid: 22699504). This study shown that the Dll4 blockage inhibits atherosclerosis and metabolic disorders (Fukuda et al., 2012; pmid: 22699504). This clone was also utilized in flow cytometry to analyse the expression of Dll4 on splenocytes, dendritic cells and macrophages (Moriyama et al., 2008; pmid: 18381350; Sekine et al., 2009; 19181931). Further, it was also used in the immunohistochemistry to stain frozen murine spleen samples (Sekine et al., 2009; 19181931). It was also used as a part of a control mixture in the functional studies on the maintenance of CD8- dendritic cells (Sekine et al., 2009; 19181931).
