- Application Supplier NoteThe binding affinity of this antibody towards native and denatured recombinant E1E2 lysate was assessed using ELISA. The binding characterization of this antibody to glycoprotein E2 of HCV was analyzed by flow cytometry using the scFv version of this antibody. Further to assess the ability of this antibody to neutralize HCV a FFU assay was performed against the J6/JFH1 recombinant virus using the human IgG1 version of this antibody. The human IgG1 version was found to neutralized HCVcc of genotypes 1a, 4a and 6a however 212.1.1 could not neutralize cell-free J6 HCVcc. Finally alanine scanning was used to mutate the E2 glycoproteins. These mutated proteins were then used to map the epitope of the human version of this antibody (Keck et al, 2019; pmid:31100098).
