- Application Supplier NoteThe original mouse IgM antibody was developed and tested to specifically recognize Abeta bound to GM1 (GM1-Abeta). A subsequent version, 4396C, an IgG2a subtype chimeric antibody, was generated and evaluated. The chimeric antibody retained its specificity for GM1-Abeta, as demonstrated by electron microscopy (EM), where it specifically labeled GM1-containing liposomes incubated with Abeta; Western blotting (WB), which confirmed the antibodys specificity for GM1-Abeta; a Thioflavin T (ThT) assay, which showed that the antibody inhibited amyloid fibril formation in a dose-dependent manner when incubated with GM1-containing liposomes and Abeta; and immunoelectron microscopy (IM), where it effectively bound to newly formed amyloid fibrils in the presence of synthetic Abeta and GM1-containing liposomes (US7339035B2). The chimeric antibody was assessed in three polymerization systems: (1) polymerization initiated by adding Abeta fibril fragment, (2) Abeta40 polymerization initiated from GAbeta40, and (3) Abeta42 polymerization initiated from GAbeta42. It exhibited suppression of the Abeta extension (US7741448B2).
