Hfab-aNS3 binds HCV NS3 protein and is directed against a conformational epitope, as it can bind NS3 in ELISA assays but not Western blots.

ELISA was used to demonstrate that Hfab-aNS3 binds to NS3 protein, with the relative affinity found to be in the range of 1 nmol/L (Prabhu et al, 2004). Through helicase assays it was established that Hfab-aNS3 blocks the helicase activity of NS3 protein (Prabhu et al, 2004). Intracellular expression of Hfab-aNS3 inhibits replication and expression of NS3 protein in a replicon cell line, as shown through immunocytochemistry (Prabhu et al, 2004).
