ELISA on recombinant hemagglutinnin has proven the specificity of the human version of this antibody. Further, hemagglutination inhibition assays (HAIs) were also performed to measure functional antibodies capable of inhibiting virus agglutination. For this assay the human antibody was mixed with the influenza virus to which erythrocytes were added. Then, the HAI titer was determined by the reciprocal dilution of the last well that contained non-agglutinated RBCs. Finally, human PBMCs were stained for flow cytometry using the human version of this antibody (Forgacs et al, 2021; pmid:33617543).
