    The original mouse IgG antibody binds mouse IL-1A with a binding affinity of Kd= 0.17 nM. The binding characterization of this antibody was done using ELISA. In vitro antibody potency was determined by its ability to inhibit IL-8 production by MRC-5 cells. This antibody was able to inhibit IL-8 production by MRC-5 cells with an IC50 of 2.00E-10M. This antibody was used in combination with 10G11 (anti- mouse IL-1B) antibody for the generation of anti-mouse IL-1alpha/beta DVD-Ig molecule (US7612181).

Raw264.7 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1% PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature.
