    The original mouse IgG antibody binds mouse IL-1A with a binding affinity of Kd= 0.17 nM. The binding characterization of this antibody was done using ELISA. In vitro antibody potency was determined by its ability to inhibit IL-8 production by MRC-5 cells. This antibody was able to inhibit IL-8 production by MRC-5 cells with an IC50 of 2.00E-10M. This antibody was used in combination with 10G11 (anti- mouse IL-1B) antibody for the generation of anti-mouse IL-1alpha/beta DVD-Ig molecule (US7612181).

Raw264.7 cells were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1% PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with IL-1 Alpha Polyclonal Antibody(bs-4947R)at 1:100 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2%BSA in PBS, followed by secondary antibody incubation for 40 min at room temperature.
