The binding capability of this antibody for SFTSV glycoprotein N was confirmed using ELISA and SPR. The binding affinity of this antibody for SFTSV Gn was evaluated to be Kd= 3.54X10^(-10) nM. This antibody was capable of neutralizing live virus in infected Vero cells with an IC50 of 2.805microg/mL. In an in vivo animal protection experiments the INFAR -/- mice which were injected with a dose of 10 mg/kg of SF5 antibody showed a survival rate of 100%. This antibody in combination with another anti-SFTSV Gn antibody SF5 was used for the generation of a bispecific antibody. In vivo experiments with bispecific antibody increased the survival rates of animals to 100% (CN114736291; PMID:28096333).
