The antibody reacts with native and denatured large T antigen. It does not react with small T antigen of SV40 or large T antigen of BKV and human papovavirus.

The simian virus 40 large T antigen plays important role in SV40 transfection and transformation of the cells. It binds to Rb and p53 suppressor proteins leading to the inactivation of suppressor pathway.
