    Monoclonal Antibody. Native human TIE-2 in ELISA experiments and on the surface of different human cell types. Isotype: Mouse IgG1. Clone: tek9. Applications: ELISA, FACS, WB. Lyophilized. TIE-2 acts as cell-surface receptor for ANGPT1, ANGPT2 and ANGPT4 and regulates angiogenesis, endothelial cell survival, proliferation, migration, adhesion and cell spreading, reorganization of the actin cytoskeleton, but also maintenance of vascular quiescence. It has anti-inflammatory effects by preventing the leakage of proinflammatory plasma proteins and leukocytes from blood vessels. It is required for normal angiogenesis and heart development during embryogenesis and for post-natal hematopoiesis. After birth, it activates or inhibits angiogenesis, depending on the context. It inhibits angiogenesis and promotes vascular stability in quiescent vessels, where endothelial cells have tight contacts. HUVEC cells(black) were fixed with 4% PFA for 10min at room temperature,permeabilized with 0.1% PBST for 20 min at room temperature, and incubated in 5% BSA blocking buffer for 30 min at room temperature. Cells were then stained with _x000D_ TIE2/CD202b Polyclonal Antibody(bs-1300R)at 1:50 dilution in blocking buffer and incubated for 30 min at room temperature, washed twice with 2% BSA in PBS, followed by secondary antibody(blue) incubation for 40 min at room temperature. Acquisitions of 20,000 events were performed. Cells stained with primary antibody (green), and isotype control (orange).

Figure 1. Western blot analysis of TEK using anti-TEK antibody (A01274-1). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human placenta tissue lysates, Lane 2: human Caco-2 whole cell lysates, Lane 3: human Hela whole cell lysates, Lane 4: rat liver tissue lysates, Lane 5: mouse liver tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TEK antigen affinity purified polyclonal antibody (Catalog # A01274-1) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TEK at approximately 160KD. The expected band size for TEK is at 126KD.
