- Scientific DescriptionTranscription termination factor 1(TTF-1) is an essential protein that controls the RNA Polymerase I transcription of the rDNA genes. TTF1 binds to the Sal Box, a repetitive sequence element in the spacer region downstream of the 3’ ends of the genes. Binding of TTF1 to this element can pause the elongation complex through the activity of chromatin remodeling complexes, such as NoRC. TTF1 also has an activating function by binding an upstream terminator element to induce chromatin remodeling. TTF1 localization can be controlled by key tumor suppressor ARF, which binds to the TTF1 nucleolar localization sequence, sending it to the nucleoplasm. This process drastically reduces rRNA synthesis. Furthermore, TTF1 can be ubiquitinylated by MDM2, which targets it for degradation. TTF1 has also been identified to play a role in HIV-1 replication.
