The CHEK2 protein is a serine/threonine protein kinase that critically coordinates checkpoint-mediated cell cycle arrest, DNA repair activation, and apoptosis in response to DNA double-strand breaks. Activated CHEK2 phosphorylates CDC25A, CDC25B, and CDC25C, inhibiting their phosphatase activities and preventing the CDK-cyclin complex from activating cell cycle progression.
