Gambogic amide acts as a potent and selective agonist for TrkA, promoting tyrosine phosphorylation and activating downstream signaling pathways such as Akt and MAPK. It specifically binds to the cytoplasmic juxtamembrane domain of the TrkA receptor, stimulating dimerization and activation.
