GGTase-I (Geranylgeranyltransferase-I) catalyzes the transfer of the farnesyl and geranylgeranyl groups from farnesyl and geranylgeranyl-diphosphate to proteins containing a C-terminal CaaX motif, where C is a conserved cysteine that is the site of farnesyl modification, a is usually an aliphatic amino acid, and X is leucine or phenylalanine. Farnesyltransferase (FT) and GGTase-I are closely related, sharing a common alpha subunit and 30% identity in their beta subunits. The GST-Tag facilitates the proteins application in typical GST pull-down assays.
