  • Scientific Description
    HtrA3, also known as PRSP (pregnancy-related serine protease), is a 453 amino acid secreted protein that contains one PDZ domain, one Kazal-like domain and one IGFBP N-terminal domain. Widely expressed with particularly high expression in ovary, heart, placenta and bladder, HtrA3 functions as a probable serine protease that is thought to play an important role in placental development. HtrA3 is downregulated in ovarian tumors, suggesting an alternative role in tumor suppression. HtrA3 exists as two alternatively spliced isoforms, designated short and long. The gene encoding HtrA3 maps to human chromosome 4, which encodes nearly 6% of the human genome and has the largest gene deserts (regions of the genome with no protein encoding genes) of all of the human chromosomes.

Figure 1. Western blot analysis of HtrA3 using anti-HtrA3 antibody (A05478). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: PANC-1 whole Cell lysates, Lane 2: HEPG2 whole cell lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-HtrA3 antigen affinity purified polyclonal antibody (Catalog # A05478) at 0.5 microg/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for HtrA3 at approximately 49KD. The expected band size for HtrA3 is at 49KD.
