Immunohistochemistry: LS-B3023 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3023 was determined to be 2 ug/ml. Peptide ELISA: antibody detection limit dilution 1:64000. Western Blot: Preliminary experiments gave an approx 150kD band in Human Cerebellum lysates after 0.1 ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the band we observe given the calculated size of 57.7kD according to NP_065105.2. The 150kD band was successfully blocked by incubation with the immunizing peptide..
