KBM 501 (Medium for Activation of Human NK Cells) Overview This product is a culture medium suitable for activating human lymphocytes (T cells, NK cells, dendritic cells). Cytokines appropriate for the target cells should be added at the time of use. Add 5 to 10% serum (plasma) at the start of activation culture. It is recommended to use KBM 502 and KBM 502B for the expansion culture after activation. This product contains recombinant human interleukin-2 (IL-2) at 1750 JRU/mL (2810 IU/mL). Osmolality: 290 +/- 15 mOsm/kg H2O (determined by freezing point depression osmometry) pH: 7.2 +/- 0.2 (determined using a glass electrode) Sterilization: Negative (Membrane filtration) Mycoplasma Test: Negative (PCR detection) Endotoxin Test: Less than 0.3 EU/mL (Determined by Limulus amebocyte lysate assay) Notice: This product is only for research use, and not for human or animal therapeutic use. Proliferative test: The culture was started at a seeding densityof27x104 cells/mL, under static conditions at 37°C,5%CO2. The culture was diluted by adding each medium containing plasma upon cell proliferation. At day10,cells unstained by trypan blue were counted. Analysis: At day10,surface markers of activated lymphocytes were analyzed byflowcytometry,andcytotoxicitytoK562cells were measured with TeraScan that is a micro fluoro cytometer developed for measuring cytotoxicity.
