  • Scientific Description
    p35 / nck5a, is a neuron-specific activator of Cdk 5. Treatment of cultured neurons with cyclohexamide induces the rapid disappearance of p35 accompanied by parallel inactivation of the kinase activity of Cdk 5. The disappearance of p35 is blocked with proteasome inhibitors, indicating the involvement of proteasome. The degradation of p35 is induced with okadaic acid in the presence of ATP in neuron extracts. The degradation of p35 by proteasome in cultured neurons is stimulated by okadaic acid in the absence of cyclohexamide. Cdk 5 and its neuron specific regulator p35 are essential for neuronal migration and for the laminar configuration of the cerebral cortex. p35 / Cdk 5 kinase concentrates at the leading edges of axonal growth cones and regulates neurite outgrowth in cortical neurons in culture. p35 / Cdk 5 kinase co-localizes with Rac in neuronal growth cones.
