- Antibody SpecificityThis confirmation-dependent antibody is specific for human INSR, reacting with an epitope within the Fn0 domain of the alpha subunit (amino acids 485-592). It cross reacts weakly with rabbit INSR, but not bovine, porcine, ovine, rat or mouse INSR.
- Application Supplier NoteThis antibody has been used in immunoprecipitation assays, with radiolabelled human insulin receptor (hINSR) and hINSR-radiolabelled insulin complexes (Soos et al, 1986), and a dimeric soluble hINSR fragment containing the first four domains of the receptor (amino acids 1-593) (Surinya et al, 2002). It has also been used in immunoblot analysis with purified hINSR (Soos et al, 1986), revealing that it binds to the receptors alpha subunit. This antibody has been shown to cause >80% inhibition of insulin binding to its receptor, in assays with IM-9 cells (Soos et al, 1986), human adipocytes (Taylor et al, 1987) and NIH 3T3 fibroblasts transfected with hINSR cDNA (Soos et al, 1989). Additionally, this antibody has been shown to mimic the metabolic effects of insulin in serum-starved cells, stimulating 2-deoxyglucose uptake to 90% of the maximal insulin effect at 100nM, and thymidine incorporation to 30% of the maximal insulin effect (Soos et al, 1989). This insulin-mimetic antibody has also been shown to stimulate receptor kinase activity and receptor tyrosine phosphorylation in intact cells (Steele-Perkins et al, 1990; Brindle et al, 1990).
