- Application Supplier NoteThe original format of this antibody (mouse IgG1) showed binding activity against E.coli-expressed recombinant human IL-13 in a sandwich ELISA with an EC50 of 0.049 and 0.039 microg/ml for human and cynomolgus IL-13. The antibody also exhibited potent binding and neutralization activities against various forms of IL-13. In inhibition assays, the antibody showed IC50 values of 0.165 microg/ml for blocking human IL-13 binding to IL-13Ralpha1 and 0.056 microg/ml for IL-13 binding to IL-13Ralpha2. Moreover, it demonstrated strong neutralization capabilities with ND50 (neutralizing dose) values of 0.0783 microg/ml for human IL-13 and 0.04 microg/ml for cynomolgus IL-13, indicating its efficacy in inhibiting IL-13-induced TF-1 cell proliferation (where the ND50 value is the concentration of monoclonal antibody required to reduce TF-1 cell proliferation by 50%, in response to a set concentration of IL-13). Furthermore, the antibody effectively neutralized mammalian-expressed human IL-13 with an ND 50 value of 0.037 microg/ml and a recombinant Q130 human IL-13 variant with an ND50 value of 0.110 microg/ml. The binding affinity of the antibody was measured by SPR and found to be 32, 270, and 61.2 pM for det-1 tagged human IL-13, human IL-13, and cynomolgus IL-13, respectively (EP1802658B1).
