The original mouse IgG antibody binds human IL-1B with a binding affinity of Kd= 0.55 nM. The binding characterization of this antibody was done using ELISA. In vitro antibody potency was determined by its ability to inhibit IL-8 production by MRC-5 cells. This antibody was able to inhibit IL-8 production by MRC-5 cells with an IC50 of 3.20E-10M. This antibody was used in combination with 6H3.1A4 (anti-IL-1alpha) antibody for the generation of anti-human IL-1alpha/beta DVD-Ig molecule that had therapeutic potential (PMID: 20068402).
